Application Notes for anti-IGF-II antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of normal human placenta. The recommended concentration ranges from 0.5 ug/ml-5.0 ug/ml. An HRP-labeled polymer detection system was used with DAB chromogen. Recommended antigen retrievals are heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer or enzyme induced antigen retrieval with Proteinase K.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of hIGF-II (10 ng/ml), a concentration of 3.1 - 4.6 ug/ml of this antibody is required.

Sandwich ELISA: To detect hIGF-II by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human IGF-II as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hIGF-II.

Western Blot: To detect hIGF-II by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIGF-II is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-IGF-II antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IGF-II

This gene encodes a member of the insulin family of polypeptide growth factors, which are involved in development and growth. It is an imprinted gene, expressed only from the paternal allele, and epigenetic changes at this locus are associated with Wilms tumour, Beckwith-Wiedemann syndrome, rhabdomyosarcoma, and Silver-Russell syndrome. A read-through INS-IGF2 gene exists, whose 5' region overlaps the INS gene and the 3' region overlaps this gene. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2010]

UniProt Comments for IGF-II

IGF2: The insulin-like growth factors possess growth-promoting activity. In vitro, they are potent mitogens for cultured cells. IGF-II is influenced by placental lactogen and may play a role in fetal development. Epigenetic changes of DNA hypomethylation in IGF2 are a cause of Silver-Russell syndrome (SRS). A clinically heterogeneous condition characterized by severe intrauterine growth retardation, poor postnatal growth, craniofacial features such as a triangular shaped face and a broad forehead, body asymmetry, and a variety of minor malformations. The phenotypic expression changes during childhood and adolescence, with the facial features and asymmetry usually becoming more subtle with age. Belongs to the insulin family. 2 isoforms of the human protein are produced by alternative splicing.
