Application Notes for anti-TGF- alpha antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive ductal carcinoma. The recommended concentration is 0.5 mug/ml with a two-hour incubation at room temperature. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of Human TGF- alpha (0.5 ng/ml), a concentration of 2.5-3.7 ug/ml of this antibody is required.

Sandwich ELISA: To detect Human TGF- alpha by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human TGF- alpha as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human TGF- alpha.

Western Blot: To detect Human TGF- alpha by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant Human TGF- alpha is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-TGF- alpha antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for TGF- alpha

This gene encodes a growth factor that is a ligand for the epidermal growth factor receptor, which activates a signaling pathway for cell proliferation, differentiation and development. This protein may act as either a transmembrane-bound ligand or a soluble ligand. This gene has been associated with many types of cancers, and it may also be involved in some cases of cleft lip/palate. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011]

UniProt Comments for TGF- alpha

TGFalpha: TGF alpha is a mitogenic polypeptide that is able to bind to the EGF receptor/EGFR and to act synergistically with TGF beta to promote anchorage-independent cell proliferation in soft agar. Interacts with the PDZ domains of MAGI3, SDCBP and SNTA1. The interaction with SDCBP, is required for the targeting to the cell surface. In the endoplasmic reticulum, in its immature form (i.e. with a prosegment and lacking full N-glycosylation), interacts with CNIH. In the Golgi apparatus, may form a complex with CNIH and GORASP2. Interacts (via cytoplasmic C-terminal domain) with NKD2. Isoform 1, isoform 3 and isoform 4 are expressed in keratinocytes and tumor-derived cell lines. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Motility/polarity/chemotaxis; Ligand, receptor tyrosine kinase; Membrane protein, integral; Cell cycle regulation

Chromosomal Location of Human Ortholog: 2p13

Cellular Component: extracellular space; cell surface; perinuclear region of cytoplasm; basolateral plasma membrane; integral to plasma membrane; plasma membrane; cytoplasmic vesicle; nucleus

Molecular Function: protein binding; growth factor activity; glycoprotein binding; epidermal growth factor receptor binding

Biological Process: epidermal growth factor receptor signaling pathway; response to drug; cell proliferation; positive regulation of epidermal growth factor receptor activity; wound healing; positive regulation of mitosis; activation of MAPK activity; angiogenesis; positive regulation of epithelial cell proliferation; negative regulation of apoptosis
