Application Notes for anti-SERPINI1 antibody
Direct Detection: To detect Human Neuroserpin by direct ELISA (using 100 ml/well) a concentration of approximately 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human Neuroserpin.

Sandwich ELISA: To detect Human Neuroserpin by sandwich ELISA (using 100 ml/well) a concentration of 0.25 - 1.0 mug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti- Human Neuroserpin as a capture antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human Neuroserpin.

Western Blot: To detect Human Neuroserpin by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 mg/ml. When used in conjunction with compatible development reagents the detection limit for recombinant Human Neuroserpin is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-SERPINI1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for SERPINI1

This gene encodes a member of the serpin superfamily of serine proteinase inhibitors. The protein is primarily secreted by axons in the brain, and preferentially reacts with and inhibits tissue-type plasminogen activator. It is thought to play a role in the regulation of axonal growth and the development of synaptic plasticity. Mutations in this gene result in familial encephalopathy with neuroserpin inclusion bodies (FENIB), which is a dominantly inherited form of familial encephalopathy and epilepsy characterized by the accumulation of mutant neuroserpin polymers. Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Jul 2008]

UniProt Comments for SERPINI1

SERPINI1: Serine protease inhibitor that inhibits plasminogen activators and plasmin but not thrombin. May be involved in the formation or reorganization of synaptic connections as well as for synaptic plasticity in the adult nervous system. May protect neurons from cell damage by tissue-type plasminogen activator. Defects in SERPINI1 are the cause of familial encephalopathy with neuroserpin inclusion bodies (FEN1B). FEN1B is characterized clinically as an autosomal dominantly inherited dementia, histologically by unique neuronal inclusion bodies and biochemically by polymers of neuroserpin. Belongs to the serpin family.
