Product Overview

Online Datasheet in PDF
Product Name
Epidermal Growth Factor Receptor 2 Extracellular Domain (HER 2 ECD), ELISA Kit
Full Product Name
Human Epidermal Growth Factor Receptor 2 Extracellular Domain ELISA Kit
Product Gene Name
Species Reactivity
Human
Assay Type
Quantitative Competitive or Sandwich
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Sensitivity
1.0 ng/mL
Specificity
This assay has high sensitivity and excellent specificity for detection of EGFR2ED. No significant cross-reactivity or interference between EGFR2ED and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between EGFR2ED and all the analogues, therefore, cross reaction may still exist in some cases.
Preparation and Storage
Store all reagents at 2-8 degree C.
Molecular Weight
97,253 Da
OMIM
613659
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
ISO Certification
Manufactured in an ISO 13485:2016 Certified Laboratory.

Other Notes

Small volumes of HER 2 ECD elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS727062 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Epidermal Growth Factor Receptor 2 Extracellular Domain (HER 2 ECD) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing HER 2 ECD. The ELISA analytical biochemical technique of the MBS727062 kit is based on HER 2 ECD antibody-HER 2 ECD antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect HER 2 ECD antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, HER 2 ECD. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

NCBI Summary for HER 2 ECD

This gene encodes a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. This protein has no ligand binding domain of its own and therefore cannot bind growth factors. However, it does bind tightly to other ligand-bound EGF receptor family members to form a heterodimer, stabilizing ligand binding and enhancing kinase-mediated activation of downstream signalling pathways, such as those involving mitogen-activated protein kinase and phosphatidylinositol-3 kinase. Allelic variations at amino acid positions 654 and 655 of isoform a (positions 624 and 625 of isoform b) have been reported, with the most common allele, Ile654/Ile655, shown here. Amplification and/or overexpression of this gene has been reported in numerous cancers, including breast and ovarian tumors. Alternative splicing results in several additional transcript variants, some encoding different isoforms and others that have not been fully characterized. [provided by RefSeq, Jul 2008]

UniProt Comments for HER 2 ECD

HER2: a proto-oncogenic receptor tyrosine kinase of the EGFR family. Essential component of a neuregulin-receptor complex, although neuregulins do not interact with it alone. Not activated by EGF, TGF- alpha and amphiregulin. Amplified in breast cancer. Overexpression induces constitutive activity, and the gene is amplified or overexpressed in up to 30% of breast cancers, correlating with poor survival. The antibody Herceptin is approved for treatment of metastatic breast cancer with HER2 amplification/overexpression. Somatic mutations seen in 4% of lung cancers and also in breast, gastric, ovarian cancer and glioblastoma. One SNP shows predisposition to breast and gastric cancer. Inhibitors: Herceptin, lapatinib, PKI-166, EKB-569, CI-1033.
