Western Blot (WB) of anti-AKT2 antibody

Western blot analysis of AKT2 expression in HeLa (A), HepG2 (B), NIH3T3 (C), mouse brain (D), rat brain (E) whole cell lysates.
Rabbit Anti-AKT2 Polyclonal Antibody – Cat# MBS820210

Immunohistochemistry (IHC) of anti-AKT2 antibody

Immunohistochemical analysis of AKT2 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-AKT2 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS820210

Immunofluorescence (IF) of anti-AKT2 antibody

Immunofluorescent analysis of AKT2 staining in HepG2 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Other Notes

Small volumes of anti-AKT2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for AKT2

This gene is a putative oncogene encoding a protein belonging to a subfamily of serine/threonine kinases containing SH2-like (Src homology 2-like) domains. The gene was shown to be amplified and overexpressed in 2 of 8 ovarian carcinoma cell lines and 2 of 15 primary ovarian tumors. Overexpression contributes to the malignant phenotype of a subset of human ductal pancreatic cancers. The encoded protein is a general protein kinase capable of phophorylating several known proteins. UniProt Comments for AKT2

Akt2: an AGC kinase. Plays critical roles in glucose metabolism and the development or maintenance of proper adipose tissue and islet mass for which other Akt/PKB isoforms are unable to fully compensate. Amplified and overexpressed in human ovarian carcinoma cell lines and amplified in some primary ovarian and pancreatic tumors. Antisense blocks invasiveness in xenografts. Expressed in several insulin-responsive tissues, and one case of Type II diabetes has been associated with a likely LOF point mutation. Mouse mutants have defects in insulin response.
