Western Blot (WB) of anti-HIST1H1B antibody

Western blot analysis of Histone H1 expression in HepG2 (A), Raw264.7 (B), PC12 (C) whole cell lysates.
Rabbit Anti-HIST1H1B Polyclonal Antibody – Cat# MBS820271

Immunohistochemistry (IHC) of anti-HIST1H1B antibody

Immunohistochemical analysis of Histone H1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-HIST1H1B Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS820271

Immunofluorescence (IF) of anti-HIST1H1B antibody

Immunofluorescent analysis of Histone H1 staining in Raw264.7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

NCBI Summary for HIST1H1B

Histones are basic nuclear proteins responsible for nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a member of the histone H1 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the small histone gene cluster on chromosome 6p22-p21.3. [provided by RefSeq, Jul 2008]

UniProt Comments for HIST1H1B

H1B: Histone H1 protein binds to linker DNA between nucleosomes forming the macromolecular structure known as the chromatin fiber. Histones H1 are necessary for the condensation of nucleosome chains into higher-order structured fibers. Acts also as a regulator of individual gene transcription through chromatin remodeling, nucleosome spacing and DNA methylation. Belongs to the histone H1/H5 family.

Protein type: DNA-binding

Chromosomal Location of Human Ortholog: 6p22.1

Cellular Component: nuclear chromatin; nucleosome; nuclear heterochromatin

Molecular Function: chromatin DNA binding; histone deacetylase binding

Biological Process: positive regulation of histone H3-K9 methylation; nucleosome assembly; establishment and/or maintenance of chromatin architecture; protein stabilization; negative regulation of transcription from RNA polymerase II promoter; positive regulation of cell growth
