Western Blot (WB) of anti-NR2F6 antibody

Western blot analysis of TEL expression in CCRFCEM (A), Jurkat (B), K562 (C) whole cell lysates.
Rabbit Anti-NR2F6 Polyclonal Antibody – Cat# MBS820690

Immunohistochemistry (IHC) of anti-NR2F6 antibody

Immunohistochemical analysis of TEL staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-NR2F6 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS820690

Immunofluorescence (IF) of anti-NR2F6 antibody

Immunofluorescent analysis of TEL staining in Jurkat cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Other Notes

Small volumes of anti-NR2F6 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for NR2F6

EAR-2: Transcription factor predominantly involved in transcriptional repression. Binds to promoter/enhancer response elements that contain the imperfect 5'-AGGTCA-3' direct or inverted repeats with various spacings which are also recognized by other nuclear hormone receptors. Involved in modulation of hormonal responses. Represses transcriptional activity of the lutropin-choriogonadotropic hormone receptor/LHCGR gene, the renin/REN gene and the oxytocin-neurophysin/OXT gene. Represses the triiodothyronine-dependent and -independent transcriptional activity of the thyroid hormone receptor gene in a cell type- specific manner. The corepressing function towards thyroid hormone receptor beta/THRB involves at least in part the inhibition of THRB binding to triiodothyronine response elements (TREs) by NR2F6. Inhibits NFATC transcription factor DNA binding and subsequently its transcriptional activity. Acts as transcriptional repressor of IL-17 expression in Th-17 differentiated CD4(+) T cells and may be involved in induction and/or maintenance of peripheral immunological tolerance and autoimmunity. Involved in development of forebrain circadian clock; is required early in the development of the locus coeruleus (LC). Belongs to the nuclear hormone receptor family. NR2 subfamily.

Protein type: Nuclear receptor; DNA-binding

Chromosomal Location of Human Ortholog: 19p13.1

Cellular Component: nucleoplasm; nucleus

Molecular Function: ligand-dependent nuclear receptor activity; protein binding; DNA binding; zinc ion binding; sequence-specific DNA binding; steroid hormone receptor activity; thyroid hormone receptor activity; transcription factor activity

Biological Process: transcription initiation from RNA polymerase II promoter; intracellular receptor-mediated signaling pathway; neuron development; gene expression; steroid hormone mediated signaling; negative regulation of transcription from RNA polymerase II promoter; detection of temperature stimulus involved in sensory perception of pain; signal transduction; entrainment of circadian clock by photoperiod
