Western Blot (WB) of anti-RAD23B antibody

Western blot analysis of RB1 expression in Jurkat (A), COLO205 (B), HuvEc (C), A431 (D), NIH3T3 (E) whole cell lysates.
Rabbit Anti-RAD23B Polyclonal Antibody – Cat# MBS820850

Immunohistochemistry (IHC) of anti-RAD23B antibody

Immunohistochemical analysis of RB1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-RAD23B Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS820850

Immunofluorescence (IF) of anti-RAD23B antibody

Immunofluorescent analysis of RB1 staining in COLO205 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

NCBI Summary for RAD23B

The protein encoded by this gene is one of two human homologs of Saccharomyces cerevisiae Rad23, a protein involved in the nucleotide excision repair (NER). This protein was found to be a component of the protein complex that specifically complements the NER defect of xeroderma pigmentosum group C (XP-c) cell extracts in vitro. This protein was also shown to interact with, and elevate the nucleotide excision activity of 3-methyladenine-DNA glycosylase (MPG), which suggested a role in DNA damage recognition in base excision repair. This protein contains an N-terminal ubiquitin-like domain, which was reported to interact with 26S proteasome, and thus this protein may be involved in the ubiquitin mediated proteolytic pathway in cells. Alternative splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Sep 2011]

UniProt Comments for RAD23B

RAD23B: Multiubiquitin chain receptor involved in modulation of proteasomal degradation. Binds to polyubiquitin chains. Proposed to be capable to bind simultaneously to the 26S proteasome and to polyubiquitinated substrates and to deliver ubiquitinated proteins to the proteasome. May play a role in endoplasmic reticulum- associated degradation (ERAD) of misfolded glycoproteins by association with PNGase and delivering deglycosylated proteins to the proteasome. Belongs to the RAD23 family.

Protein type: Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 9q31.2

Cellular Component: proteasome complex; nucleoplasm; cytoplasm; nucleus

Molecular Function: protein binding; damaged DNA binding; polyubiquitin binding; single-stranded DNA binding

Biological Process: nucleotide-excision repair, DNA damage recognition; proteasomal ubiquitin-dependent protein catabolic process; nucleotide-excision repair; regulation of proteasomal ubiquitin-dependent protein catabolic process; spermatogenesis; nucleotide-excision repair, DNA damage removal; DNA repair
