Western Blot (WB) of anti-EIF4G2 antibody

Western blot analysis of EIF4G2 expression in MCF7 (A), HeLa (B), K562 (C), Jurkat (D), HepG2 (E) whole cell lysates.
Rabbit Anti-EIF4G2 Polyclonal Antibody – Cat# MBS820897

Immunohistochemistry (IHC) of anti-EIF4G2 antibody

Immunohistochemical analysis of EIF4G2 staining in human liver cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescence (IF) of anti-EIF4G2 antibody

Immunofluorescent analysis of EIF4G2 staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

NCBI Summary for EIF4G2

Translation initiation is mediated by specific recognition of the cap structure by eukaryotic translation initiation factor 4F (eIF4F), which is a cap binding protein complex that consists of three subunits: eIF4A, eIF4E and eIF4G. The protein encoded by this gene shares similarity with the C-terminal region of eIF4G that contains the binding sites for eIF4A and eIF3; eIF4G, in addition, contains a binding site for eIF4E at the N-terminus. Unlike eIF4G, which supports cap-dependent and independent translation, this gene product functions as a general repressor of translation by forming translationally inactive complexes. In vitro and in vivo studies indicate that translation of this mRNA initiates exclusively at a non-AUG (GUG) codon. Alternatively spliced transcript variants encoding different isoforms of this gene have been described. [provided by RefSeq, Jul 2008]

UniProt Comments for EIF4G2

EIF4G2: a general repressor of translation by forming translationally inactive complexes. Appears to play a role in the switch from cap-dependent to IRES-mediated translation during mitosis, apoptosis and viral infection. Interacts with the protein kinases Mnk1 and -2. Binds EIF4A and EIF3. Cleaved by some caspases and viral proteases.
