Western Blot (WB) of anti-TNFAIP8 antibody

Western blot analysis of SCC-S2 expression in K562 (A), Raw264.7 (B) whole cell lysates.
Rabbit Anti-TNFAIP8 Polyclonal Antibody – Cat# MBS821136

Immunohistochemistry (IHC) of anti-TNFAIP8 antibody

Immunohistochemical analysis of SCC-S2 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-TNFAIP8 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS821136

Immunofluorescence (IF) of anti-TNFAIP8 antibody

Immunofluorescent analysis of SCC-S2 staining in Raw264.7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Other Notes

Small volumes of anti-TNFAIP8 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for TNFAIP8

TNFAIP8: Acts as a negative mediator of apoptosis and may play a role in tumor progression. Suppresses the TNF-mediated apoptosis by inhibiting caspase-8 activity but not the processing of procaspase-8, subsequently resulting in inhibition of BID cleavage and caspase-3 activation. Belongs to the TNFAIP8 family. 3 isoforms of the human protein are produced by alternative splicing.

Chromosomal Location of Human Ortholog: 5q23.1

Cellular Component: cytoplasm

Molecular Function: caspase inhibitor activity

Biological Process: defense response to Gram-positive bacterium; interleukin-1 beta production; negative regulation of caspase activity; negative regulation of apoptosis
