Western Blot (WB) of anti-TOP2A antibody

Western blot analysis of Topoisomerase 2 alpha expression in MDAMB468 (A), MDAMB231 (B), HeLa (C) whole cell lysates.
Rabbit Anti-TOP2A Polyclonal Antibody – Cat# MBS821529

Immunohistochemistry (IHC) of anti-TOP2A antibody

Immunohistochemical analysis of Topoisomerase 2 alpha staining in human liver cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-TOP2A Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS821529

Immunofluorescence (IF) of anti-TOP2A antibody

Immunofluorescent analysis of Topoisomerase 2 alpha staining in MDAMB231 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Western Blot (WB) of anti-TOP2A antibody

Western blot analysis of Topoisomerase 2 alpha expression in Hela (A), H446 (B) whole cell lysates.
Rabbit Anti-TOP2A Polyclonal Antibody, Western Blot (WB) – Cat# MBS821529

Other Notes

Small volumes of anti-TOP2A antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for TOP2A

This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromosome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia. [provided by RefSeq, Jul 2010]
