Western Blot (WB) of anti-BRF1 antibody

Western blot analysis of BRF1 expression in Jurkat (A), NIH3T3 (B) whole cell lysates.
Rabbit Anti-BRF1 Polyclonal Antibody – Cat# MBS822292

Immunohistochemistry (IHC) of anti-BRF1 antibody

Immunohistochemical analysis of BRF1 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-BRF1 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS822292

Immunofluorescence (IF) of anti-BRF1 antibody

Immunofluorescent analysis of BRF1 staining in NIH3T3 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Other Notes

Small volumes of anti-BRF1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for BRF1

This gene encodes one of the three subunits of the RNA polymerase III transcription factor complex. This complex plays a central role in transcription initiation by RNA polymerase III on genes encoding tRNA, 5S rRNA, and other small structural RNAs. The gene product belongs to the TF2B family. Several alternatively spliced variants encoding different isoforms, that function at different promoters transcribed by RNA polymerase III, have been identified. [provided by RefSeq, Jun 2011]

UniProt Comments for BRF1

GTF3B: General activator of RNA polymerase which utilizes different TFIIIB complexes at structurally distinct promoters. The isoform 1 is involved in the transcription of tRNA, adenovirus VA1, 7SL and 5S RNA. Isoform 2 is required for transcription of the U6 promoter. Belongs to the TFIIB family. 7 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription initiation complex

Chromosomal Location of Human Ortholog: 14q

Cellular Component: nucleoplasm; transcription factor TFIIIB complex

Molecular Function: zinc ion binding; TATA-binding protein binding

Biological Process: transcription from RNA polymerase III promoter; transcription initiation from RNA polymerase III promoter; tRNA transcription; regulation of transcription, DNA-dependent; gene expression; regulation of mRNA stability; rRNA transcription
