Western Blot (WB) of anti-ATP1A1 antibody

Western blot analysis of ATP1A1 expression in HeLa (A), A549 (B), HepG2 (C) whole cell lysates.
Rabbit Anti-ATP1A1 Polyclonal Antibody – Cat# MBS827440

Immunohistochemistry (IHC) of anti-ATP1A1 antibody

Immunohistochemical analysis of ATP1A1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescence (IF) of anti-ATP1A1 antibody

Immunofluorescent analysis of ATP1A1 staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

NCBI Summary for ATP1A1

The protein encoded by this gene belongs to the family of P-type cation transport ATPases, and to the subfamily of Na+/K+ -ATPases. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The catalytic subunit of Na+/K+ -ATPase is encoded by multiple genes. This gene encodes an alpha 1 subunit. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May 2009]

UniProt Comments for ATP1A1

ATP1A1: the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of Na and K ions across the plasma membrane. This action creates the electrochemical gradient of Na and K, providing the energy for active transport of various nutrients. Two splice-variant isoforms have been described.
