Western Blot (WB)

Application Notes for anti-DKK2 antibody

Western blotting (0.5-4 ug/ml). However, the optimal conditions should be determined individually. The antibody recognizes 30-35 kDa Dkk2 in samples from human, mouse and rat origins. A ~25 kDa band can also be detected, presumably to be the cleavage fragment of Dkk2.

Small volumes of anti-DKK2 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. DKK2: Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero- posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease. Interacts with LRP5 and LRP6. Expressed in heart, brain, skeletal muscle and lung. Belongs to the dickkopf family.
