Persephin (PSPN) Active Protein

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS844158

Background: Persephin is a disulfide-linked homodimer neurotrophic factor structurally related to GDNF, Artemin, and Neurturin. These proteins belong to the cysteine-knot family of growth factors that assume stable dimeric structures. Persephin signals… More details

Solubility/Reconstitution Instructions
Centrifuge the vial prior to opening. Reconstitute in 5 mM Sodium Phosphate, pH 7.5 to a concentration of <= 0.5 mg/ml. Do not vortex. This solution can be stored at 2-8 degree C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20 degree C to -80 degree C.
Endotoxin Level
< 0.1 ng/mug of protein (<1EU/mug).
Activity (Specifications/test method)
Human Persephin induces RET phosphorylation using a concentration range of 0.1-1.0 ng/ml. Human Persephin binds to mammalian GFRalpha4 with the Kd of 100pM. Other members of the GDNF family (Artemin, GDNF and Neurturin) do not bind to mammalian GFRalpha4.
Biological Activity

Handling
Centrifuge the vial prior to opening.
Applications Tested/Suitable for PSPN active protein
SDS-PAGE; HPLC

Other Notes

Small volumes of PSPN active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PSPN

The protein encoded by this gene is a neurotrophic factor, belonging to the GDNF family. Neurotrophic factors are important for the proper development and maintenance of the nervous system. These factors promote neuronal survival and can prevent the neuronal degeneration associated with injury, toxin exposure, or neurodegenerative disease. The encoded protein has amino acid similarity to its other family members, glial cell line-derived neurotrophic factor and neurturin. This gene product promotes the survival of ventral midbrain dopaminergic neurons in culture and prevents their degeneration after 6-hydroxydopamine treatment in vivo. [provided by RefSeq, Jul 2008]
