Solubility/Reconstitution Instructions
Centrifuge the vial prior to opening. Reconstitute in sterile PBS, pH 7.4 to a concentration of 50 ug/ml. Do not vortex. This solution can be stored at 2-8 degree C for up to 1 month. For extended storage, it is recommended to store at -20 degree C.
Endotoxin Level
<1 EU/mug by LAL method
Biological Activity
Measured by its binding ability in a functional ELISA. Immobilized rh PD-L1 / B7-H1 at 2 mug/ml (100 mul/well) can bind rhPD1/PDCD1 Fc Chimera with a linear range of 1-10 mug/ml. Measured by its ability to inhibit anti-CD3 antibody induced IL-2 secretion in human T lymphocytes. The ED50 for this effect is typically 0.6-5 mug/ml.

UniProt Comments for CD274

CD274: Involved in the costimulatory signal, essential for T- cell proliferation and production of IL10 and IFNG, in an IL2- dependent and a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation and cytokine production. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing.
