Other Notes

Small volumes of FMR1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS931448 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the fragile X mental retardation 1 (FMR1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing FMR1. The ELISA analytical biochemical technique of the MBS931448 kit is based on FMR1 antibody-FMR1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FMR1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FMR1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%). Fragile X syndrome is an X-linked genetic disease that results in developmental problems including learning and cognitive disabilities. Mutations in the FMR1 gene can result in fragile X syndrome. The causative mutation is almost always an expansion of a CGG triplet repeat, from a normal 5-40 repeats to 200-1000 repeats. This silences the FMR1 gene, resulting in the lack of Fragile X mental retardation 1 protein (FMRP) expression. FMRP is normally expressed in brain, testes, and ovarian tissues. In the brain, this protein is thought to play a role in the development of synaptic plasticity. FMRP has been found to act as a mRNA shuttle, carrying mRNA molecules from the nucleus to the cytoplasm or localizing mRNA to the dendrites, and regulating protein synthesis at synapses. FMRP not only binds to mRNA, but it also seems to interact with ribosomes, inhibiting translation.

This Human Fragile X mental retardation 1 protein, FMR1 Elisa kit is highly specific for human FMRP and has been optimized for the detection of FMRP in serum, plasma, and tissue homogenates.

NCBI Summary for FMR1

The protein encoded by this gene binds RNA and is associated with polysomes. The encoded protein may be involved in mRNA trafficking from the nucleus to the cytoplasm. A trinucleotide repeat (CGG) in the 5' UTR is normally found at 6-53 copies, but an expansion to 55-230 repeats is the cause of fragile X syndrome. Expansion of the trinucleotide repeat may also cause one form of premature ovarian failure (POF1). Multiple alternatively spliced transcript variants that encode different protein isoforms and which are located in different cellular locations have been described for this gene. UniProt Comments for FMR1

FMR1: Translation repressor. Component of the CYFIP1-EIF4E- FMR1 complex which binds to the mRNA cap and mediates translational repression. In the CYFIP1-EIF4E-FMR1 complex this subunit mediates translation repression. RNA- binding protein that plays a role in intracellular RNA transport and in the regulation of translation of target mRNAs. Associated with polysomes. May play a role in the transport of mRNA from the nucleus to the cytoplasm. Binds strongly to poly(G), binds moderately to poly(U) but shows very little binding to poly(A) or poly(C). Component of the CYFIP1-EIF4E-FMR1 complex which is composed of CYFIP, EIF4E and FMR1. Interacts with CYFIP1 and CYFIP2. The interaction with brain cytoplasmic RNA 1 (BC1) increases binding affinity for the CYFIP1-EIF4E complex in the brain. Homooligomer. Found in a RNP granule complex with IGF2BP1. Directly interacts with SMN and TDRD3. Interacts with the SMN core complex that contains SMN1, GEMIN2/SIP1, DDX20/GEMIN3, GEMIN4, GEMIN5, GEMIN6, GEMIN7, GEMIN8 and STRAP/UNRIP. Interacts with FXR1, FXR2, IGF2BP1, NUFIP1, NUFIP2, MCRS1 and RANBP9. Highest levels found in neurons, brain, testis, placenta and lymphocytes. Also expressed in epithelial tissues and at very low levels in glial cells. Belongs to the FMR1 family. 8 isoforms of the human protein are produced by alternative splicing.
