ELISA KitBone morphogenetic proteins (BMPs) belong to a group of growth and differentiation factors called transforming growth factors (TGFs). BMPs play an important part during development and tissue regeneration. BMPs, more specifically BMP, are being increasingly used in orthopaedic surgery due to advanced research into osteoinductive factors that may enhance and improve bone therapy. Fractures and critical size bone defects (defects that do not heal without intervention) can frequently occur as a result of trauma and disease, and their management requires mediation. There are many areas in therapy that BMP is being applied to, including dental treatment, open tibial fractures, cancer and spinal surgery. Since its discovery in 1988, more than 20 different types of BMPs have been uncovered. Through preclinical testing, the use of BMPs has been extensively investigated, and BMP_2 has been introduced as one of the postpotent BMPs.
Although BMP is used clinically for skeletal repair, in vivo administration can induce abnormal, adipose-filled, poor-quality bone. The differentiation factor NEL-like molecule-1 (NELL-1) has been reported as osteoinductive in multiple in vivo preclinical models. A study demonstrated that NELL-1 combined with BMP2 significantly optimizes osteogenesis in a rodent femoral segmental defect model by minimizing the formation of BMP2-induced adipose-filled cystlike bone. The studies establish the feasibility of combining NELL-1 with BMP2 to improve clinical bone regeneration and provide mechanistic insight into canonical Wnt pathway activity during NELL-1 and BMP2 osteogenesis. The novel abilities of NELL-1 to stimulate Wnt signaling and to repress adipogenesis may highlight new treatment approaches for bone loss in osteoporosis.
