The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Gly-Gl n-Asp-Arg.
Host
E Coli
Form/Format
Supplied as a lyophilized powder without additives. Reconstitute with sterile dH2O, 0.1% HSA or BSA to 0.1mg/ml, which can then be further diluted to other aqueous solutions.
Purity/Purification
Highly Purified
95% as determined by RP-HPLC, anion-exchange FPLC and/or reducing and non-reducing SDS-PAGE Silver stained gel. Endotoxin: 0.1ng/ug (IEU/ug).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile dH2O, 0.1% HSA or BSA,,aliquot and store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
The ED50=1-10ng/ml as determined by the dose-dependent proliferation of human peripheral blood mononuclear cells is < 5ng/ml.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.54 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of IL-21 as a Reference Standard.

Other Notes

Small volumes of IL21 active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL21

This gene encodes a member of the common-gamma chain family of cytokines with immunoregulatory activity. The encoded protein plays a role in both the innate and adaptive immune responses by inducing the differentiation, proliferation and activity of multiple target cells including macrophages, natural killer cells, B cells and cytotoxic T cells. Dysregulation of this gene plays a role in multiple immune-mediated diseases including lupus, psoriasis and chronic inflammatory diseases. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Dec 2011]

UniProt Comments for IL21

Function: Cytokine with immunoregulatory activity. May promote the transition between innate and adaptive immunity. Induces the production of IgG1 and IgG3 in B-cells

By similarity. May play a role in proliferation and maturation of natural killer (NK) cells in synergy with IL15. May regulate proliferation of mature B- and T-cells in response to activating stimuli. In synergy with IL15 and IL18 stimulates interferon gamma production in T-cells and NK cells. During T-cell mediated immune response may inhibit dendritic cells (DC) activation and maturation. Ref.1 Ref.6

Subcellular location: Secreted Ref.1.

Tissue specificity: Expressed in activated CD4-positive T-cells but not in CD8-positive T-cells, B-cells, or monocytes. Ref.1

Sequence similarities: Belongs to the IL-15/IL-21 family.

Sequence caution: The sequence AAG29348.1 differs from that shown. Reason: Erroneous initiation. The sequence AAH66258.1 differs from that shown. Reason: Erroneous initiation.
