Application Notes for anti-VPS29 antibody
Peptide ELISA: Antibody detection limit dilution 1: 16000.
Immunohistochemistry: In paraffin embedded Human Uterus shows vesicular and membranous staining in the endometrial glands. Recommended concentration, 4-8ug/ml.
Western Blot: Preliminary experiments gave an approx 60kDa band in Human Spleen lysates after 0.3ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 20.5kDa according to NP_057310.1 and 20.9kDa according to NP_476528.1. The 60kDa band was successfully blocked by incubation with the immunizing peptide.

Immunohistochemistry (IHC) of anti-VPS29 antibody

(3.8ug/ml) staining of paraffin embedded Human Uterus. Steamed antigen retrieval with citrate buffer pH 6, AP-staining.

Other Notes

Small volumes of anti-VPS29 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for VPS29

This gene belongs to a group of vacuolar protein sorting (VPS) genes that, when functionally impaired, disrupt the efficient delivery of vacuolar hydrolases. The protein encoded by this gene is a component of a large multimeric complex, termed the retromer complex, which is involved in retrograde transport of proteins from endosomes to the trans-Golgi network. This VPS protein may be involved in the formation of the inner shell of the retromer coat for retrograde vesicles leaving the prevacuolar compartment. Alternative splice variants encoding different isoforms and representing non-protein coding transcripts have been found for this gene. [provided by RefSeq, Aug 2013]

UniProt Comments for VPS29

VPS29: Essential component of the retromer complex, a complex required to retrieve lysosomal enzyme receptors (IGF2R and M6PR) from endosomes to the trans-Golgi network. Also required to regulate transcytosis of the polymeric immunoglobulin receptor (pIgR-pIgA). Has low protein phosphatase activity towards a serine-phosphorylated peptide derived from IGF2R (in vitro). Belongs to the VPS29 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Vesicle; EC 3.1.3.3; Protein phosphatase, Ser/Thr (non-receptor)

Chromosomal Location of Human Ortholog: 12q24

Cellular Component: cytoplasm; cytosol; early endosome; endosome; endosome membrane; intracellular membrane-bound organelle; late endosome; retromer complex

Molecular Function: metal ion binding; protein binding; protein transporter activity

Biological Process: intracellular protein transport; retrograde transport, endosome to Golgi; Wnt receptor signaling pathway
