Specificity
CD6 is a type I transmembrane glycoprotein that contains a 24-amino acid signal sequence, three extracellular -scavenger receptor cysteine-rich- (SRCR) domains, a membrane-spanning domain and a 44-amino acid cytoplasmic domain. The CD6 glycoprotein is tyrosine phosphorylated during TCR-mediated T cell activation. CD6 shows significant homology to CD5. CD6 is present on mature thymocytes, peripheral T cells and a subset of B cells. Antibodies to CD6 are used to deplete T cells from bone marrow transplants to prevent graft versus host disease.

Flow Cytometry (0.5-1ug/million cells in 0.1ml)
Immunofluorescence (0.5-1ug/ml)
Immunohistochemistry (Formalin-fixed) (0.5-1ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate Buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes)

Immunohistochemistry (IHC) of anti-CD6 antibody

Formalin-fixed, paraffin-embedded human Tonsil stained with CD6 Monoclonal Antibody (C6/372+3F7B5).

Other Notes

Small volumes of anti-CD6 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CD6

This gene encodes a protein found on the outer membrane of T-lymphocytes as well as some other immune cells. The encoded protein contains three scavenger receptor cysteine-rich (SRCR) domains and a binding site for an activated leukocyte cell adhesion molecule. The gene product is important for continuation of T cell activation. This gene may be associated with susceptibility to multiple sclerosis (PMID: 19525953, 21849685). Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Dec 2011]

UniProt Comments for CD6

CD6: a type I membrane protein involved in cell adhesion. Binds to CD166. Expressed on thymocytes, peripheral T cells, a subset of B cells, and a subset of neurons. Mediates the binding of developing thymocytes with thymic epithelial cells. CD6- T cells are less autoreactive than CD6+ T cells. Phosphorylated on tyrosines and serines following T cell antigen receptor signaling. Five splice-variant isoforms have been described.

Protein type: Cell surface; Membrane protein, integral

Chromosomal Location of Human Ortholog: 11q13

Cellular Component: immunological synapse; integral to plasma membrane; intrinsic to plasma membrane; T cell receptor complex

Molecular Function: lipopolysaccharide binding; protein binding
