PDS1 Antibody – Mouse Monoclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS601156

Pds1 is an anaphase inhibitor and plays important role in DNA damage and spindle check point pathways. Pds1 inhibits sister chromatid separation by binding and inhibiting Esp1, cystein protease that causes cleavage of the cohesin Scc1 that binds the sister… More details

Application Notes for anti-PDS1 antibody
Dilution: Western Blotting (Not verified), Immunohistochemistry (formalin/paraffin) (Ab 1-2ug/ml for 30 min at RT) [Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.] The optimal dilution for a specific application should be determined by the investigator.

Immunohistochemistry (IHC) of anti-PDS1 antibody

Formalin-fixed, paraffin-embedded human tonsil stained with Pds1 Ab using peroxidase-conjugate and AEC chromogen. Note nuclear staining of lympocytes in germinal center.

Other Notes

Small volumes of anti-PDS1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for PDS1

Function: Regulatory protein, which plays a central role in chromosome stability. Probably acts by blocking the action of key proteins. During the mitosis, it blocks Separase/ESP1 function, preventing the proteolysis of the cohesin complex and the subsequent segregation of the chromosomes. At the onset of anaphase, it is ubiquitinated, conducting to its destruction and to the liberation of ESP1. Ref.6

Subunit structure: Interacts with the caspase-like ESP1, and prevents its protease activity probably by covering its active site. Interacts with CDC20. Ref.6 Ref.7

Subcellular location: Cytoplasm. Nucleus.

Domain: The N-terminal destruction box (D-box) acts as a recognition signal for degradation via the ubiquitin-proteasome pathway

By similarity.

Post-translational modification: Phosphorylated by CDC28. The phosphorylation may be important for ESP1 localization to the nucleus. Ref.8Ubiquitinated by the anaphase promoting complex (APC) at the onset of anaphase, conducting to its degradation. Ref.5

Sequence similarities: Belongs to the securin family.
