Application Notes for anti-CLEC9A antibody
Suitable for use in Western Blot and Immunohistochemistry.
Dilution: Western Blot: 1ug/ml
Immunohistochemistry: 5-15ug/ml

Western Blot (WB) of anti-CLEC9A antibody

Western blot analysis of lysate of human CD14+ peripheral blood mononuclear cells (PBMC). PVDF Membrane was probed with 1ug/ml of MBS647676 followed by HRP-conjugated sheep IgG secondary antibody. A specific band was detected for CLEC9a at approximately 45kD. This experiment was conducted under reducing conditions. The following buffers were used: blotting buffer: 25mM Tris, pH 7.4, 0.5M NaCl and 0.05% Tween-20; blocking solution: 3% BSA in blotting buffer, pH adjusted to 7.4; antibody solution: 2% dry non-fat dry milk in blotting buffer, pH adjusted to 7.4.

Immunohistochemistry (IHC) of anti-CLEC9A antibody

IHC staining of immersion fixed, paraffin-embedded sections of human spleen using MBS647676 at 15ug/ml overnight at 4°C. Tissue was stained using the anti-sheep HRP-DAB Cell & Tissue Staining Kit and counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm.

NCBI Summary for CLEC9A

CLEC9A is a group V C-type lectin-like receptor (CTLR) that functions as an activation receptor and is expressed on myeloid lineage cells (Huysamen et al., 2008 [PubMed 18408006]).[supplied by OMIM, Aug 2008]

UniProt Comments for CLEC9A

Function: Functions as an endocytic receptor on a small subset of myeloid cells specialized for the uptake and processing of material from dead cells. Recognizes filamentous form of actin in association with particular actin-binding domains of cytoskeletal proteins, including spectrin, exposed when cell membranes are damaged, and mediate the cross-presentation of dead-cell associated antigens in a Syk-dependent manner. Ref.3 Ref.4

Subunit structure: Homodimer. Ref.1 Ref.4

Subcellular location: Membrane; Single-pass type II membrane protein Ref.1.

Tissue specificity: In peripheral blood highly restricted on the surface of BDCA31+ dendritic cells and on a small subset of CD14+ and CD16- monocytes. Ref.1 Ref.3

Post-translational modification: N-glycosylated. Ref.1

Sequence similarities: Contains 1 C-type lectin domain.
