25,243 Da
Molecular Structure
A soluble fusion protein consisting of the extracellular (134aa) domain of human CD28 fused to murine IgG2a Fc (233aa).
Transfectant Cell Line
CHO
Performance
CD28-muIg was tested for activity and quantitated in EIA. Fusion protein was captured using Goat-anti-mouse antibody coated plate. After 45 minute incubation, plates were washed twice and an excess of Mouse Ig was added to completely block the plate. Anti-CD28/Biotin followed by Streptavidin/HRP, and TMB/H2O2 were used for detection. Fusion protein was detectable at 4 ng/ml in this assay.
Production
Human CD28-muIg fusion protein was purified from (low FBS containing) tissue culture supernatant of CHO transfectants by Protein A and size exclusion chromatography.
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 0.5 mg/ml Gentamicin Sulfate (as a preservative).
Group Header
CD28-muIg

UniProt Comments for CD28

CD28: a type I plasma membrane protein Involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival. Binds to B7-1 and -2. Expressed in T cells and plasma cells, but not in less mature B cells. Six splice-variant isoforms have been described.

Protein type: Cell surface; Membrane protein, integral

Cellular Component: membrane; plasma membrane; integral to membrane; immunological synapse; lipid raft; external side of plasma membrane

Molecular Function: protein homodimerization activity; SH3/SH2 adaptor activity; protease binding

Biological Process: positive regulation of isotype switching to IgG isotypes; positive regulation of mitosis; T cell activation; positive regulation of interleukin-2 biosynthetic process; negative thymic T cell selection; T cell receptor signaling pathway; positive regulation of phosphoinositide 3-kinase cascade; positive regulation of protein kinase B signaling cascade; positive regulation of inflammatory response to antigenic stimulus; T cell costimulation; immune response; positive regulation of T cell proliferation; positive regulation of transcription from RNA polymerase II promoter; positive regulation of alpha-beta T cell proliferation; regulatory T cell differentiation
