Application Notes for anti-APRIL antibody
Immunohistochemistery: This antibody stained formalin-fixed paraffin-embedded sections of human carcinoid tissue. The recommended concentration is 0.25 ug/ml with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 Sodium Citrate buffer is recommended.

Sandwich ELISA: To detect hAPRIL by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human APRIL as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hAPRIL.

Western Blot: To detect hAPRIL by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hAPRIL is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-APRIL antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for APRIL

The protein encoded by this gene is a member of the tumor necrosis factor (TNF) ligand family. This protein is a ligand for TNFRSF17/BCMA, a member of the TNF receptor family. This protein and its receptor are both found to be important for B cell development. In vitro experiments suggested that this protein may be able to induce apoptosis through its interaction with other TNF receptor family proteins such as TNFRSF6/FAS and TNFRSF14/HVEM. Alternative splicing results in multiple transcript variants. Some transcripts that skip the last exon of the upstream gene (TNFSF12) and continue into the second exon of this gene have been identified; such read-through transcripts are contained in GeneID 407977, TNFSF12-TNFSF13.
