NCBI Summary for ALKBH3

The Escherichia coli AlkB protein protects against the cytotoxicity of methylating agents by repair of the specific DNA lesions generated in single-stranded DNA. ALKBH2 (MIM 610602) and ALKBH3 are E. coli AlkB homologs that catalyze the removal of 1-methyladenine and 3-methylcytosine (Duncan et al., 2002 [PubMed 12486230]).[supplied by OMIM, Mar 2008]

UniProt Comments for ALKBH3

ALKBH3: Dioxygenase that repairs alkylated DNA containing 1- methyladenine (1meA) and 3-methylcytosine (3meC) by oxidative demethylation. Has a strong preference for single-stranded DNA. Able to process alkylated 3mC within double-stranded regions via its interaction with ASCC3, which promotes DNA unwinding to generate single-stranded substrate needed for ALKHB3. May also act on RNA. Requires molecular oxygen, alpha-ketoglutarate and iron. Belongs to the alkB family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Demethylase; Hydrolase; DNA repair, damage; Oxidoreductase; EC 1.14.11.-

Chromosomal Location of Human Ortholog: 11p11.2

Cellular Component: cytoplasm; mitochondrion; nucleoplasm; nucleus

Molecular Function: DNA demethylase activity; DNA-N1-methyladenine dioxygenase activity; ferrous iron binding; L-ascorbic acid binding; protein binding

Biological Process: cell proliferation; DNA dealkylation; DNA repair
