Western Blot (WB) of anti-PRKCQ antibody

Western blot analysis of PKC theta (pS676) expression in HeLa UV-treated (A), SP2/0 UV-treated (B), PC12 UV-treated (C) whole cell lysates.
Rabbit Anti-PRKCQ Polyclonal Antibody – Cat# MBS820626

Immunohistochemistry (IHC) of anti-PRKCQ antibody

Immunohistochemical analysis of PKC theta (pS676) staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescence (IF) of anti-PRKCQ antibody

Immunofluorescent analysis of PKC theta (pS676) staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Other Notes

Small volumes of anti-PRKCQ antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PRKCQ

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role. The protein encoded by this gene is one of the PKC family members. It is a calcium-independent and phospholipid-dependent protein kinase. This kinase is important for T-cell activation. It is required for the activation of the transcription factors NF-kappaB and AP-1, and may link the T cell receptor (TCR) signaling complex to the activation of the transcription factors. UniProt Comments for PRKCQ

PKCT: an AGC kinase of the PKC family. A novel PKC: Ca2+ independent but still regulated by PS, DAG and phorbol esters. Contains 2 zinc-dependent phorbol-ester and DAG binding domains. Expressed in skeletal muscle and hematopoietic cells. May play a role in T cell receptor signaling and insulin resistance. Required for TCR-induced NF-kappaB activation in mature T lymphocytes.
