Interleukin 1 alpha, Recombinant, Human (IL-1a, Hematopoietin-1, Lymphocyte-activating Factor, LAF, Endogenous Pyrogen, Leukocyte Endogenous Mediator, LEM, Mononuclear Cell Factor, MCF)
Product Gene Name
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Ser-Ala-Pr o-Phe-Ser.
Host
E Coli
Form/Format
Supplied as a lyophilized powder in 20mM Tris-HCl, pH 8.0, 5mM MgCl2, 10% glycerol.
Purity/Purification
Highly Purified
98% by RP-HPLC, FPLC, or reducing/non-reducing SDS-PAGE Silver Stain. Chromatographically purified.
Preparation and Storage
Lyophilized powder is stable for at least 12 months at -20 degree C Reconstituted product must be stored in aliquots at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Biological Activity
rHuIL-1alpha is fully biologically active when compared to standards. The ED50 as determined by the dose-dependent stimulation of murine D10S cells is less then 0.001ng/ml, corresponding to a Specific Activity of 1 x 10e6 IU/mg.
Dimers and Aggregates
1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug)
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280nm using the absorbency value of 1.13 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of chicken IL-1alpha as a Reference Standard.
Reconstitution
Reconstitute the lyophilized rHuIL-1alpha in sterile 18M -cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.

Other Notes

Small volumes of IL1A active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for IL1A

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is a pleiotropic cytokine involved in various immune responses, inflammatory processes, and hematopoiesis. This cytokine is produced by monocytes and macrophages as a proprotein, which is proteolytically processed and released in response to cell injury, and thus induces apoptosis. This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. It has been suggested that the polymorphism of these genes is associated with rheumatoid arthritis and Alzheimer's disease. [provided by RefSeq]

UniProt Comments for IL1A

Function: Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

Subunit structure: Monomer.

Subcellular location: Secreted. Note: The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins.

Domain: The similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function.

Sequence similarities: Belongs to the IL-1 family.
