PD-L1 Antibody – Rabbit

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS488962

The binding characterization of this antibody towards human, mouse and cynomolgus monkey PD-L1 was done using flow cytometry. This antibody was shown to block the binding of hPD-1-Fc protein to hPD-L1 expressed on the surface of the HEK-293T cells at a… More details

Specificity
This antibody strongly binds human and cynomolgus monkey PD-L1 and shows weak binding for mouse PD-L1. PD-L1 is highly expressed in many human tumors.. PD-L1 exerts an immune suppressive signal through binding to PD-1 and B7.1, and tumor expression of PD-L1 can mediate cancer immune evasion. Blocking this pathway, therefore provides a rationale for cancer immunotherapy.
Preparation and Storage
Store at 4 degree C for up to 3 months. For longer storage, aliquot and store at -20 degree C.
Molecular Weight
33,275 Da
OMIM
605402
3D Structure
ModBase 3D Structure for Q9NZQ7
Animal-free Manufacturing Platform
The recombinant antibody is manufactured using a recombinant, mammalian HEK293-based expression platform. The manufacturing platform is animal-free and animal component-free.
Immunogen
The original antibody was generated by immunizing BALB/c mice with recombinant human PD-L1.
Chimeric Use Statement
This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques.

ISO Certification
Manufactured in an ISO 9001:2015 Certified Laboratory.
Applications Tested/Suitable for anti-PD-L1 antibody
Flow Cytometry (FC/FACS), Blocking (BL)

Flow Cytometry (FC/FACS) of anti-PD-L1 antibody

Flow cytometry using the Anti-PD-L1 antibody 332M7 (MBS488962). Paraformaldehyde fixed MDA-MB-231 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody or the rabbit IgG version of 332M7 (MBS488962-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunofluorescence (IF) of anti-PD-L1 antibody

Immunofluorescence staining of MDA-MB-231 cells with anti-PD-L1 (MBS488962) 332M7 Immunofluorescence analysis of paraformaldehyde fixed MDA-MB-231 cells stained with the chimeric rabbit IgG version of 332M7 (MBS488962-23.0) at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom MBS488962-23.0, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor 488 secondary antibody.

Other Notes

Small volumes of anti-PD-L1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for PD-L1

CD274: Involved in the costimulatory signal, essential for T- cell proliferation and production of IL10 and IFNG, in an IL2- dependent and a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation and cytokine production. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing.
