Form/Format
FAS protein was lyophilized from a 0.2 um filtered concentrated solution in 1xPBS, pH 7.4.
Sterile Filtered White lyophilized (freeze-dried) powder.
Purity/Purification
Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by SDS-PAGE.
Preparation and Storage
Lyophilized FAS although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution FAS should be stored at 4 degree C between 2-7 days and for future use below -18 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

12,487 Da
Active Protein Note
Protein activity can be an ambiguous term. Please refer to the data sheet or inquire with Technical Support to determine the definition of activity for this product.
Solubility
It is recommended to reconstitute the lyophilized FAS in sterile 18M-cm H2O not less than 100 ug/ml, which can then be further diluted to other aqueous solutions.
Biological Activity
The ED50 was determined by its ability to inhibit the cytotoxicity of Jurkat cells is between 10-15 ug/ml in the presence of 2ng/ml of hFasL.

Other Notes

Small volumes of FAS active protein vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for FAS

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor contains a death domain. It has been shown to play a central role in the physiological regulation of programmed cell death, and has been implicated in the pathogenesis of various malignancies and diseases of the immune system. The interaction of this receptor with its ligand allows the formation of a death-inducing signaling complex that includes Fas-associated death domain protein (FADD), caspase 8, and caspase 10. The autoproteolytic processing of the caspases in the complex triggers a downstream caspase cascade, and leads to apoptosis. This receptor has been also shown to activate NF-kappaB, MAPK3/ERK1, and MAPK8/JNK, and is found to be involved in transducing the proliferating signals in normal diploid fibroblast and T cells. Several alternatively spliced transcript variants have been described, some of which are candidates for nonsense-mediated mRNA decay (NMD). The isoforms lacking the transmembrane domain may negatively regulate the apoptosis mediated by the full length isoform. [provided by RefSeq, Mar 2011]
