Application Notes for anti-PD-L1 antibody
This antibody is capable of binding B7-H1 molecules expressed on the surface of M435 by flow cytometry. This antibody was also used for the immunohistochemical analysis of intestinal and gastric cancer tissues (WO2019085238)

Flow Cytometry (FC/FACS) of anti-PD-L1 antibody

Flow-cytometry using the anti-PD-L1 antibody XGMHc25.1 . MDA-MB-231 cells were stained with an isotype control or the rabbit-chimeric version of XGMHc25.1 (MBS489042, blue line) at a concentration of 1 µg/ml for 30 mins at RT. After washing, bound antibody was detected using Alexa Fluor 488 conjugated goat anti-rabbit IgG and cells analysed on a FACSCanto flow-cytometer.

Immunofluorescence (IF) of anti-PD-L1 antibody

Immunofluorescence staining of MDA-MB-231 cells with anti-PD-L1 XGMHc25.1 Immunofluorescence analysis of paraformaldehyde fixed MDA-MB-231 cells stained with the chimeric rabbit IgG version of XGMHc25.1 (MBS489042) (1:1000 dilution) for 1h followed by Alexa Fluor 488 secondary antibody (1:1500 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom MBS489042, DAPI, merged channels and an isotype control. The isotype control was anti-fluorescein antibody followed by staining with Alexa Fluor 488 secondary antibody.

Other Notes

Small volumes of anti-PD-L1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for PD-L1

CD274: Involved in the costimulatory signal, essential for T- cell proliferation and production of IL10 and IFNG, in an IL2- dependent and a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation and cytokine production. Belongs to the immunoglobulin superfamily. BTN/MOG family. 3 isoforms of the human protein are produced by alternative splicing.
