Function
Essential counter-regulatorycarboxypeptidase of the renin- angiotensinhormone system that is a critical regulatorof blood volume, systemic vascularresistance, and thus cardiovascularhomeostasis (PubMed:27217402). Convertsangiotensin I to angiotensin 1- 9, anine-amino acid peptide withanti-hypertrophic effects in cardiomyocytes, and angiotensin II to angiotensin 1-7, whichthen acts as a beneficial vasodilator andanti-proliferation agent, counterbalancingthe actions of the vasoconstrictorangiotensin II (PubMed:10969042,PubMed:10924499,PubMed:11815627,PubMed:19021774,PubMed:14504186). Alsoremoves the C-terminal residue from threeother vasoactive peptides, neurotensin, kinetensin, and des-Arg bradykinin, but isnot active on bradykinin (PubMed:10969042,PubMed:11815627). Alsocleaves other biological peptides, such asapelins (apelin-13, [Pyr1]apelin-13,apelin-17, apelin-36), casomorphins(beta-casomorphin- 7, neocasomorphin)and dynorphin A with high efficiency(PubMed:11815627,PubMed:27217402,PubMed:28293165). Inaddition, ACE2 C-terminus is homologous tocollectrin and is responsible for thetrafficking of the neutral amino acidtransporter SL6A19 to the plasmamembrane of gut epithelial cells via directinteraction, regulating its expression on thecell surface and its catalytic activity(PubMed:18424768,PubMed:19185582).
Cellular Location
[Processed angiotensin-converting enzyme2]: Secreted
Tissue Location
Expressed in endothelial cells from smalland large arteries, and in arterial smoothmuscle cells (at protein level)(PubMed:15141377). Expressed inenterocytes of the small intestine, Leydigcells and Sertoli cells (at protein level)(PubMed:15141377) Expressed in the renalproximal tubule and the small intestine (atprotein level) (PubMed:18424768).Expressed in heart, kidney, testis, andgastrointestinal system (at protein level)(PubMed:10969042, PubMed:10924499,PubMed:15231706, PubMed:12459472,PubMed:15671045, PubMed:32715618,PubMed:32170560). In lung, expressed atlow levels in some alveolar type 2 cells, theexpression seems to be individual- specific(at protein level) (PubMed:32425701,PubMed:15141377, PubMed:32715618,PubMed:32170560). Coexpressed withTMPRSS2 within some lung alveolar type 2cells, ileal absorptive enterocytes, intestinalepithelial cells, cornea, gallbladder andnasal goblet secretory cells(PubMed:32413319, PubMed:32327758,PubMed:32358202). Coexpressed withTMPRSS4 within mature enterocytes(PubMed:32404436)

NCBI Summary for ACE2

The protein encoded by this gene belongs to the angiotensin-converting enzyme family of dipeptidyl carboxydipeptidases and has considerable homology to human angiotensin 1 converting enzyme. This secreted protein catalyzes the cleavage of angiotensin I into angiotensin 1-9, and angiotensin II into the vasodilator angiotensin 1-7. The organ- and cell-specific expression of this gene suggests that it may play a role in the regulation of cardiovascular and renal function, as well as fertility. In addition, the encoded protein is a functional receptor for the spike glycoprotein of the human coronaviruses SARS and HCoV-NL63. [provided by RefSeq, Jul 2008]

UniProt Comments for ACE2

ACE2: Carboxypeptidase which converts angiotensin I to angiotensin 1-9, a peptide of unknown function, and angiotensin II to angiotensin 1-7, a vasodilator. Also able to hydrolyze apelin- 13 and dynorphin-13 with high efficiency. May be an important regulator of heart function. In case of human coronaviruses SARS and HCoV-NL63 infections, serve as functional receptor for the spike glycoprotein of both coronaviruses. Interacts with ITGB1. Interacts with SARS-CoV and HCoV- NL63 spike glycoprotein. Up-regulated in failing heart. Expressed in endothelial cells from small and large arteries, and in arterial smooth muscle cells. Expressed in lung alveolar epithelial cells, enterocytes of the small intestine, Leydig cells and Sertoli cells. Expressed in heart, kidney, testis, and gastrointestinal system. Activated by chloride and fluoride, but not bromide. Inhibited by MLN-4760, cFP_Leu, and EDTA, but not by the ACE inhibitors linosipril, captopril and enalaprilat. Belongs to the peptidase M2 family. 2 isoforms of the human protein are produced by alternative splicing.
