ABRAXAS Antibody – Rabbit Polyclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS3902604

Abraxas was identified in a phosphopeptide affinity proteomic and mass spec analysis as a protein that bound directly to the BRCA1 BRCT repeat. Along with RAP80, Abraxas was required to form a complex with BRCA1 for the DNA damage response. More details

Applications Tested/Suitable for anti-Abraxas antibody
Immunoprecipitation (IP), Immunohistochemistry (IHC)
Application Notes for anti-Abraxas antibody
IP: 6ug/mg lysate
IHC: 1:500-1:2000. Epitope retrieval with citrate buffer pH 6.0 is recommended for FFPE tissue sections.

Immunohistochemistry (IHC) of anti-Abraxas antibody

Detection of human Abraxas by immunohistochemistry. Sample: FFPE section of human ovarian carcinoma. Antibody: Affinity purified rabbit anti-Abraxas (Cat. No. MBS3902604) used at a dilution of 1:1,000 (1ug/ml). Detection: DAB
Rabbit Anti-ABRAXAS Polyclonal Antibody – Cat# MBS3902604

Immunoprecipitation (IP) of anti-Abraxas antibody

Detection of human Abraxas by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-Abraxas antibody MBS3902604 used for IP at 10 ug/mg lysate. Abraxas was also immunoprecipitated by rabbit anti-Abraxas antibody MBS3902603, which recognizes an upstream epitope. For blotting immunoprecipitated Abraxas, MBS3902603 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

UniProt Comments for Abraxas

Abraxas: Component of the BRCA1-A complex, a complex that specifically recognizes 'Lys-63'-linked ubiquitinated histones H2A and H2AX at DNA lesions sites, leading to target the BRCA1-BARD1 heterodimer to sites of DNA damage at double-strand breaks (DSBs). The BRCA1-A complex also possesses deubiquitinase activity that specifically removes 'Lys-63'-linked ubiquitin on histones H2A and H2AX. In the BRCA1-A complex, it acts as a central scaffold protein that assembles the various components of the BRCA1-A complex and mediates the recruitment of BRCA1. Belongs to the FAM175 family. Abraxas subfamily.
