Western Blot (WB) of anti-CUX1 antibody

CUX1 Antibody (C-term) western blot analysis in MCF-7 cell line lysates (35ug/lane).This demonstrates the CUX1 antibody detected the CUX1 protein (arrow).

Immunohistochemistry (IHC) of anti-CUX1 antibody

CUX1 Antibody (C-term) immunohistochemistry analysis in formalin fixed and paraffin embedded human kidney tissue followed by peroxidase conjugation of the secondary antibody and DAB staining. This data demonstrates the use of the CUX1 Antibody (C-term) for immunohistochemistry. Clinical relevance has not been evaluated.

Immunofluorescence (IF) of anti-CUX1 antibody

Fluorescent confocal image of Hela cell stained with CUX1 Antibody (C-term).Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with CUX1 primary antibody (1:25, 1 h at 37 degree C). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree C).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree C). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min). CUX1 immunoreactivity is localized to Nucleus significantly.

Other Notes

Small volumes of anti-CUX1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for CUX1

The protein encoded by this gene is a member of the homeodomain family of DNA binding proteins. It may regulate gene expression, morphogenesis, and differentiation and it may also play a role in the cell cycle progession. Several alternatively spliced transcript variants encoding different isoforms have been identified.[provided by RefSeq, Feb 2011]

UniProt Comments for CUX1

CUTL1: Probably has a broad role in mammalian development as a repressor of developmentally regulated gene expression. May act by preventing binding of positively-activing CCAAT factors to promoters. Component of nf-munr repressor; binds to the matrix attachment regions (MARs) (5' and 3') of the immunoglobulin heavy chain enhancer. Represses T-cell receptor (TCR) beta enhancer function by binding to MARbeta, an ATC-rich DNA sequence located upstream of the TCR beta enhancer. Belongs to the CUT homeobox family. 8 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; DNA-binding; Transcription factor

Chromosomal Location of Human Ortholog: 7q22.1

Cellular Component: integral to Golgi membrane; nucleus; cytosol

Molecular Function: sequence-specific DNA binding; chromatin binding

Biological Process: regulation of transcription from RNA polymerase II promoter; intra-Golgi vesicle-mediated transport; transcription, DNA-dependent; multicellular organismal development; auditory receptor cell differentiation; positive regulation of transcription from RNA polymerase II promoter; positive regulation of dendrite morphogenesis; negative regulation of transcription from RNA polymerase II promoter; kidney development; lung development
