Application Notes for anti-FASN antibody
WB: 1:50-1:100
FLISA: 1:1,000
Applications are based on unconjugated antibody.

Immunofluorescence (IF) of anti-FASN antibody

Fluorescent confocal image of HepG2 cells stained with FASN (Center) antibody. HepG2 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AP7449c FASN (Center) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). Note the highly specific localization of the FASN mainly to the mainly to the cytoplasm, supported by Human Protein Atlas Data.

Other Notes

Small volumes of anti-FASN antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for FASN

FASN: a multifunctional enzyme. Catalyzes the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha.
