Application Notes for anti-G-CSF antibody
Immunohistochemistery: This antibody stained formalin-fixed, paraffin-embedded sections of human angiosarcoma. The recommended concentration is 0.25 mug/mL with an overnight incubation at 4 degree C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended.

Neutralization: To yield one-half maximal inhibition [ND50] of the biological activity of hG-CSF (0.5 ng/ml), a concentration of 0.04-0.06 ug/ml of this antibody is required.

Sandwich ELISA: To detect hG-CSF by sandwich ELISA (using 100 mul/well antibody solution) a concentration of 0.5 - 2.0 mug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human G-CSF as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant hG-CSF.

Western Blot: To detect hG-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hG-CSF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Other Notes

Small volumes of anti-G-CSF antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for G-CSF

G-CSF: Granulocyte/macrophage colony-stimulating factors are cytokines that act in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. This CSF induces granulocytes. Belongs to the IL-6 superfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cytokine; Secreted, signal peptide; Secreted; Cell cycle regulation

Chromosomal Location of Human Ortholog: 17q11.2-q12

Cellular Component: extracellular space

Molecular Function: enzyme binding; growth factor activity; cytokine activity; granulocyte colony-stimulating factor receptor binding

Biological Process: positive regulation of phosphoinositide 3-kinase cascade; positive regulation of protein kinase B signaling cascade; positive regulation of peptidyl-tyrosine phosphorylation; granulocyte differentiation; positive regulation of myeloid cell differentiation; positive regulation of protein binding; positive regulation of actin filament polymerization; multicellular organismal development; cytokine and chemokine mediated signaling pathway; positive regulation of cell proliferation; positive regulation of transcription factor import into nucleus; immune response; positive regulation of transcription from RNA polymerase II promoter; positive regulation of peptidyl-serine phosphorylation
