Western Blot (WB) of anti-HSF1 antibody

Figure 1. Western blot analysis of HSF1 using anti-HSF1 antibody (MBS1754687).

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.

Lane 1: human Hela whole cell lysates,
Lane 2: human K562 whole cell lysates,
Lane 3: human MCF-7 whole cell lysates,
Lane 4: human MOLT-4 whole cell lysates,
Lane 5: human 293T whole cell lysates.

After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HSF1 antigen affinity purified polyclonal antibody (#MBS1754687) at 0.25 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for HSF1 at approximately 80 kDa. The expected band size for HSF1 is at 57 kDa.

NCBI Summary for HSF1

The product of this gene is a heat-shock transcription factor. Transcription of heat-shock genes is rapidly induced after temperature stress. Hsp90, by itself and/or associated with multichaperone complexes, is a major repressor of this gene. [provided by RefSeq, Jul 2008]

UniProt Comments for HSF1

HSF1: a transcription factor that specifically binds heat shock promoter elements (HSE) and activates transcription. Induced in response to heat, heavy metals, and oxidative stress. In higher eukaryotes, HSF is unable to bind to HSEs unless the cells are stressed. Becomes phosphorylated in response to stress, forming homotrimers that bind DNA and activate transcription. Phosphorylation by PLK1 enhances nuclear translocation, and phosphorylation by CaMKII enhances transactivation. Phosphorylation by GSK3 and ERK1 induces binding by 14-3-3 and sequestration in the cytoplasm. In addition, during attenuation from the heat shock response, HSF1 is repressed by direct binding of Hsp70, HSP40, and HSF binding protein 1 (HSBP1). Four alternatively spliced isoforms have been described.
