Immunoprecipitation (IP)
Application Notes for anti-ICT1 antibody
IP: 6ug/mg lysate

Immunoprecipitation (IP) of anti-ICT1 antibody

Detection of human ICT1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-ICT1 antibody (MBS3905942 lot 1) used for IP at 6 ug per reaction. For blotting immunoprecipitated ICT1, MBS3905943 was used at 0.4 mg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

NCBI Summary for ICT1

The protein encoded by this gene is a peptidyl-tRNA hydrolase and a vital component of the large mitochondrial ribosome. The encoded protein serves as a ribosome release factor for this ribosome, which translates mitochondrial genes. This protein may be responsible for degrading prematurely-terminated polypeptides and for reusing stalled ribosomes. Two transcript variants encoding different isoforms have been found for this gene. UniProt Comments for ICT1

ICT1: Essential peptidyl-tRNA hydrolase component of the mitochondrial large ribosomal subunit. Acts as a codon-independent translation release factor that has lost all stop codon specificity and directs the termination of translation in mitochondrion, possibly in case of abortive elongation. May be involved in the hydrolysis of peptidyl-tRNAs that have been prematurely terminated and thus in the recycling of stalled mitochondrial ribosomes. Belongs to the prokaryotic/mitochondrial release factor family. ICT1 subfamily.

Protein type: Mitochondrial; EC 3.1.1.29

Chromosomal Location of Human Ortholog: 17q25.1

Cellular Component: mitochondrion; mitochondrial matrix; mitochondrial inner membrane; mitochondrial large ribosomal subunit

Molecular Function: aminoacyl-tRNA hydrolase activity; translation release factor activity, codon nonspecific

Biological Process: mitochondrial translation; organelle organization and biogenesis
