Western Blot (WB) of anti-IKBKE antibody

Figure 1. Western blot analysis of IKKi/IKKe/IKBKE using anti-IKKi/IKKe/IKBKE antibody (MBS1753467).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: human HL-60 whole cell lysates
Lane 2: human U-87MG whole cell lysates
Lane 3: human A549 whole cell lysates.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1. 5 hour at RT. The membrane was incubated with rabbit anti-IKKi/IKKe/IKBKE antigen affinity purified polyclonal antibody (Catalog # MBS1753467) at 0. 5 μg/mL overnight at 4 degree C, then washed with TBS-0. 1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1. 5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # MBS176460) with Tanon 5200 system. A specific band was detected for IKKi/IKKe/IKBKE at approximately 80KD. The expected band size for IKKi/IKKe/IKBKE is at 80KD.

NCBI Summary for IKBKE

IKBKE is a noncanonical I-kappa-B (see MIM 164008) kinase (IKK) that is essential for regulating antiviral signaling pathways. IKBKE has also been identified as a breast cancer (MIM 114480) oncogene and is amplified and overexpressed in over 30% of breast carcinomas and breast cancer cell lines (Hutti et al., 2009 [PubMed 19481526]).[supplied by OMIM, Oct 2009]

UniProt Comments for IKBKE

IKKE: a oncogenic kinase of the IKK family. Phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. Plays a pivotal role in coordinating the activation of IRF3 and NF-kappaB in the innate immune response. Its redistribution to PML nuclear bodies upon DNA damage is TOPORS-dependent. Is a breast cancer oncogene that exerts its oncogenic properties through cRel.
