Western Blot (WB) of anti-MYST2 antibody

HBO1/MYST2 Antibody (Center) western blot analysis in Jurkat cell line lysates (35ug/lane).This demonstrates the HBO1/MYST2 antibody detected the HBO1/MYST2 protein (arrow).

Immunohistochemistry (IHC) of anti-MYST2 antibody

Formalin-fixed and paraffin-embedded human testis tissue reacted with HBO1/MYST2 antibody (Center), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.

Immunofluorescence (IF) of anti-MYST2 antibody

Fluorescent confocal image of Hela cell stained with HBO1/MYST2 Antibody (Center). Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with HBO1/MYST2 primary antibody (1:25, 1 h at 37 degree C). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree C).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree C). Nuclei were counterstained with DAPI (blue) (10 ug/ml, 10 min). HBO1/MYST2 immunoreactivity is localized to nucleus significantly.

NCBI Summary for MYST2

The protein encoded by this gene is part of the multimeric HBO1 complex, which possesses histone H4-specific acetyltransferase activity. This activity is required for functional replication origins and is involved in transcriptional activation of some genes. In both cases, the acetylation of histone H4 helps unfold chromatin so that the DNA can be accessed and replicated or transcribed. [provided by RefSeq, Oct 2016]

UniProt Comments for MYST2

MYST2: a histone acetyltransferase that participates in DNA replication and the loading of the minichromosome maintenance (Mcm) complex onto chromatin. Is required for origin recognition complex (Orc) formation and replication licensing, and is responsible for much of the acetylation of histone H4. Interacts with MCM2 and ORC1L. Phosphorylated by Cdc2 during mitosis, creating a site that recruits Plk1, which in turn phosphorylates MYST2. Its phosphorylation by Plk1 during mitosis may be required for the formation of the prereplicative complex. Interacts with the androgen receptor (AR) in the presence of dihydrotestosterone. Specifically represses AR-mediated transcription.

Protein type: EC 2.3.1.48; Transcription, coactivator/corepressor; Acetyltransferase; Nuclear receptor co-regulator
