NCBI Summary for POLB

The protein encoded by this gene is a DNA polymerase involved in base excision and repair, also called gap-filling DNA synthesis. The encoded protein, acting as a monomer, is normally found in the cytoplasm, but it translocates to the nucleus upon DNA damage. Several transcript variants of this gene exist, but the full-length nature of only one has been described to date. [provided by RefSeq, Sep 2011]

UniProt Comments for POLB

POLB: Repair polymerase that plays a key role in base-excision repair. Has 5'-deoxyribose-5-phosphate lyase (dRP lyase) activity that removes the 5' sugar phosphate and also acts as a DNA polymerase that adds one nucleotide to the 3' end of the arising single-nucleotide gap. Conducts 'gap-filling' DNA synthesis in a stepwise distributive fashion rather than in a processive fashion as for other DNA polymerases. Monomer. Interacts with APEX1, HUWE1/ARF-BP1, STUB1/CHIP and USP47. Belongs to the DNA polymerase type-X family.

Protein type: DNA repair, damage; Transferase; EC 2.7.7.7; DNA replication; Lyase

Chromosomal Location of Human Ortholog: 8p11.2

Cellular Component: cytoplasm; microtubule; nucleoplasm; nucleus; protein complex; spindle microtubule

Molecular Function: damaged DNA binding; DNA-directed DNA polymerase activity; enzyme binding; lyase activity; metal ion binding; microtubule binding; protein binding

Biological Process: aging; base-excision repair; base-excision repair, gap-filling; DNA damage response, signal transduction resulting in induction of apoptosis; DNA repair; DNA-dependent DNA replication; homeostasis of number of cells; inflammatory response; lymph node development; neuron apoptosis; nucleotide-excision repair, DNA gap filling; pyrimidine dimer repair; response to DNA damage stimulus; response to ethanol; response to gamma radiation; response to hyperoxia; salivary gland morphogenesis; somatic hypermutation of immunoglobulin genes; spleen development
