Western Blot (WB) of anti-PTGR1 antibody

Figure 1. Western blot analysis of PTGR1 using anti-PTGR1 antibody (MBS1758935).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: human HepG2 whole cell lysates,
Lane 3: human A549 whole cell lysates,
Lane 4: human RT4 whole cell lysates,
Lane 5: human SiHa whole cell lysates,
Lane 6: human Hacat whole cell lysates,
Lane 7: human Caco-2 whole cell lysates,
Lane 8: human PC-3 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PTGR1 antigen affinity purified polyclonal antibody (#MBS1758935) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (#MBS176460) with Tanon 5200 system. A specific band was detected for PTGR1 at approximately 36 kDa. The expected band size for PTGR1 is at 36 kDa.

Flow Cytometry (FC/FACS) of anti-PTGR1 antibody

Figure 2. Flow Cytometry analysis of RT4 cells using anti-PTGR1 antibody (MBS1758935).
Overlay histogram showing RT4 cells stained with MBS1758935 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-PTGR1 Antibody (MBS1758935, 1ug/1x106 cells) for 30 min at 20 degree C. DyLight488 conjugated goat anti-rabbit IgG (MBS177405, 5-10ug/1x106 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

NCBI Summary for PTGR1

This gene encodes an enzyme that is involved in the inactivation of the chemotactic factor, leukotriene B4. The encoded protein specifically catalyzes the NADP+ dependent conversion of leukotriene B4 to 12-oxo-leukotriene B4. A pseudogene of this gene is found on chromosome 1. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Mar 2009]

UniProt Comments for PTGR1

LTB4DH: Functions as 15-oxo-prostaglandin 13-reductase and acts on 15-oxo-PGE1, 15-oxo-PGE2 and 15-oxo-PGE2-alpha. Has no activity towards PGE1, PGE2 and PGE2-alpha. Catalyzes the conversion of leukotriene B4 into its biologically less active metabolite, 12-oxo-leukotriene B4. This is an initial and key step of metabolic inactivation of leukotriene B4. Belongs to the NADP-dependent oxidoreductase L4BD family.

Protein type: Oxidoreductase; EC 1.3.1.74; EC 1.3.1.48

Chromosomal Location of Human Ortholog: 9q31.3

Cellular Component: cytoplasm

Molecular Function: 15-oxoprostaglandin 13-oxidase activity; zinc ion binding; 2-alkenal reductase activity

Biological Process: response to toxin; cyclooxygenase pathway; arachidonic acid metabolic process; leukotriene metabolic process
