Western Blot (WB) of anti-HEXIM1 antibody

Western blot analysis of HEXIM1 expression in A431 (A), Jurkat (B), NIH3T3 (C), rat heart (D) whole cell lysates.
Anti-HEXIM1 Polyclonal Antibody – Cat# MBS8243164

Immunohistochemistry (IHC) of anti-HEXIM1 antibody

Immunohistochemical analysis of HEXIM1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Anti-HEXIM1 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS8243164

Immunofluorescence (IF) of anti-HEXIM1 antibody

Immunofluorescent analysis of HEXIM1 staining in A431 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
Anti-HEXIM1 Polyclonal Antibody, Immunofluorescence (IF) – Cat# MBS8243164

Other Notes

Small volumes of anti-HEXIM1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for HEXIM1

Expression of this gene is induced by hexamethylene-bis-acetamide in vascular smooth muscle cells. This gene has no introns. [provided by RefSeq, Jul 2008]

UniProt Comments for HEXIM1

HEXIM1: Transcriptional regulator which functions as a general RNA polymerase II transcription inhibitor. In cooperation with 7SK snRNA sequesters P-TEFb in a large inactive 7SK snRNP complex preventing RNA polymerase II phosphorylation and subsequent transcriptional elongation. May also regulate NF-kappa-B, ESR1, NR3C1 and CIITA-dependent transcriptional activity. Homooligomer and heterooligomer with HEXIM2; probably dimeric. Component of the 7SK snRNP complex at least composed of P-TEFb (composed of CDK9 and CCNT1/cyclin-T1), HEXIM1, HEXIM2, BCDIN3, SART3 proteins and 7SK and U6 snRNAs. Interacts with the N-CoR complex through NCOR1. Interacts with ESR1 and NR3C1. May interact with NF-kappa-B through RELA. Up-regulated by HMBA (hexamethylene bisacetamide). Down-regulated by estrogen. Ubiquitously expressed with higher expression in placenta. HEXIM1 and HEXIM2 are differentially expressed. Expressed in endocrine tissues. Belongs to the HEXIM family.

Protein type: Transcription factor; Motility/polarity/chemotaxis

Chromosomal Location of Human Ortholog: 17q21.31

Cellular Component: cytoplasm; nucleoplasm; nucleus
