Product Categories/Family for anti-ATG5 antibody

Applications Tested/Suitable for anti-ATG5 antibody
Immunofluorescence (IF), FLISA, Immunohistochemistry (IHC), Western Blot (WB)
Application Notes for anti-ATG5 antibody
IF: 1:10-50
FLISA: 1:1,000
IHC: 1:50-100
WB: 1:100-500
Applications are based on unconjugated antibody.

Immunofluorescence (IF) of anti-ATG5 antibody

Fluorescent image of U251 cells stained with ATG5 (N-term) antibody. U251 cells were treated with Chloroquine (50 uM,16h), then fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AP1812a ATG5 (N-term) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). ATG5 immunoreactivity is localized to autophagic vacuoles in the cytoplasm of U251 cells.

Western Blot (WB) of anti-ATG5 antibody

APG5L Antibody (D3) western blot analysis in Hela cell line lysates (35ug/lane).This demonstrates the APG5L antibody detected the APG5L protein (arrow).
Rabbit Anti-ATG5 Polyclonal Antibody, Western Blot (WB) – Cat# MBS6276200

Western Blot (WB) of anti-ATG5 antibody

The anti-APG5L Pab is used in Western blot to detect APG5L in mouse brain tissue lysate.
Rabbit Anti-ATG5 Polyclonal Antibody, Western Blot (WB) – Cat# MBS6276200

Western Blot (WB) of anti-ATG5 antibody

Western blot analysis of APG5L (arrow) using rabbit polyclonal APG5L Antibody (D3). 293 cell lysates (2 ug/lane) either nontransfected (Lane 1) or transiently transfected (Lane 2) with the APG5L gene.
Rabbit Anti-ATG5 Polyclonal Antibody, Western Blot (WB) – Cat# MBS6276200

Immunohistochemistry (IHC) of anti-ATG5 antibody

Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.

Other Notes

Small volumes of anti-ATG5 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for ATG5

ATG5: Required for autophagy. Conjugates to ATG12 and associates with isolation membrane to form cup-shaped isolation membrane and autophagosome. The conjugate detaches from the membrane immediately before or after autophagosome formation is completed. The ATG5-ATG12 conjugate forms a complex with several units of ATG16. Interacts with TECPR1; the interaction is direct and does not take place when ATG16 is associated with the ATG5- ATG12 conjugate. By apoptotic stimuli. Ubiquitous. The mRNA is present at similar levels in viable and apoptotic cells, whereas the protein is dramatically highly expressed in apoptotic cells. Belongs to the ATG5 family. 2 isoforms of the human protein are produced by alternative splicing.
