Applications Tested/Suitable for anti-MITF antibody
Western Blot (WB), Immunohistochemistry (IHC)

Wester Blot (WB) of anti-MITF antibody

Dilution: WB (1/500 - 1/1000), IH (1/50 - 1/100)
Western blot analysis of MITF (pS180) expression in U87MG (A), A549 (B) whole cell lysates.
Rabbit Anti-MITF Polyclonal Antibody – Cat# MBS9240636

Immunohistochemistry (IHC) of anti-MITF antibody

Dilution: WB: (1/500 - 1/1000), IH (1/50 - 1/100)
Immunohistochemical analysis of MITF (pS180) staining in human heart formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

ELISA (EIA) of anti-MITF antibody

Dilution: WB (1/500 - 1/1000), IH (1/50 - 1/100)
Rabbit Anti-MITF Polyclonal Antibody, ELISA (EIA) – Cat# MBS9240636

Other Notes

Small volumes of anti-MITF antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for MITF

This gene encodes a transcription factor that contains both basic helix-loop-helix and leucine zipper structural features. It regulates the differentiation and development of melanocytes retinal pigment epithelium and is also responsible for pigment cell-specific transcription of the melanogenesis enzyme genes. Heterozygous mutations in the this gene cause auditory-pigmentary syndromes, such as Waardenburg syndrome type 2 and Tietz syndrome. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]

UniProt Comments for MITF

MITF: a transcription factor that contains both basic helix-loop-helix and leucine zipper structural features. Plays a critical role in the differentiation of various cell types including neural crest- derived melanocytes, mast cells, osteoclasts and optic cup-derived retinal pigment epithelium. Two isoforms are known: the M-isoform is expressed exclusively in melanocytes, while the A-isoform has a much broader range of expression. Mutations in MITF can lead to Waardenburg syndrome. Ten alternatively spliced isoforms have been described.

Protein type: DNA-binding; Transcription factor; Oncoprotein

Chromosomal Location of Human Ortholog: 3p14.2-p14.1

Cellular Component: protein complex; nucleus
