Western Blot (WB) of anti-ATIC antibody

Western blot analysis of ATIC expression in MCF7 (A), HepG2 (B), HT29 (C), K562 (D) whole cell lysates.
Rabbit Anti-ATIC Polyclonal Antibody – Cat# MBS8245700

Immunohistochemistry (IHC) of anti-ATIC antibody

Immunohistochemical analysis of ATIC staining in rat brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-ATIC Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS8245700

Immunofluorescence (IF) of anti-ATIC antibody

Immunofluorescent analysis of ATIC staining in U2OS cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Other Notes

Small volumes of anti-ATIC antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for ATIC

This gene encodes a bifunctional protein that catalyzes the last two steps of the de novo purine biosynthetic pathway. The N-terminal domain has phosphoribosylaminoimidazolecarboxamide formyltransferase activity, and the C-terminal domain has IMP cyclohydrolase activity. A mutation in this gene results in AICA-ribosiduria. [provided by RefSeq, Sep 2009]

UniProt Comments for ATIC

ATIC: Bifunctional enzyme that catalyzes 2 steps in purine biosynthesis. Defects in ATIC are the cause of AICAR transformylase/IMP cyclohydrolase deficiency (AICAR). A neurologically devastating inborn error of purine biosynthesis. Patients excrete massive amounts of AICA-riboside in the urine and accumulate AICA- ribotide and its derivatives in erythrocytes and fibroblasts. AICAR causes profound mental retardation, epilepsy, dysmorphic features and congenital blindness. Belongs to the PurH family.

Protein type: Cofactor and Vitamin Metabolism - one carbon pool by folate; EC 3.5.4.10; Methyltransferase; Nucleotide Metabolism - purine; EC 2.1.2.3; Hydrolase

Chromosomal Location of Human Ortholog: 2q35

Cellular Component: cytosol; membrane; mitochondrion

Molecular Function: IMP cyclohydrolase activity; phosphoribosylaminoimidazolecarboxamide formyltransferase activity; protein homodimerization activity

Biological Process: 'de novo' IMP biosynthetic process; cerebellum development; cerebral cortex development; dihydrofolate metabolic process; nucleobase, nucleoside, nucleotide and nucleic acid metabolic process; nucleoside metabolic process; organ regeneration; purine ribonucleoside monophosphate biosynthetic process; response to inorganic substance; tetrahydrofolate biosynthetic process
