Applications Tested/Suitable for anti-CPI17 antibody
Western Blot (WB), Immunofluorescence (IF), Immunohistochemistry (IHC)

Wester Blot (WB) of anti-CPI17 antibody

Dilution: WB 1/500 - 1/1000
Western blot analysis of CPI17 expression in A2780 (A), HEK293T (B), EC9706 (C), mouse brain (D), rat brain (E) whole cell lysates.
Rabbit Anti-CPI17 Polyclonal Antibody – Cat# MBS9233500

Immunohistochemistry (IHC) of anti-CPI17 antibody

Dilution: IH 1/50 - 1/200
Immunohistochemical analysis of CPI17 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Rabbit Anti-CPI17 Polyclonal Antibody, Immunohistochemistry (IHC) – Cat# MBS9233500

Immunofluorescence (IF) of anti-CPI17 antibody

Dilution: IF: 1/50 - 1/200
Immunofluorescent analysis of CPI17 staining in Jurkat cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 degree C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

NCBI Summary for CPI17

The protein encoded by this gene belongs to the protein phosphatase 1 (PP1) inhibitor family. This protein is an inhibitor of smooth muscle myosin phosphatase, and has higher inhibitory activity when phosphorylated. Inhibition of myosin phosphatase leads to increased myosin phosphorylation and enhanced smooth muscle contraction. Alternatively spliced transcript variants encoding different isoforms have been noted for this gene. [provided by RefSeq, Sep 2011]

UniProt Comments for CPI17

PPP1R14A: a phosphorylation-dependent inhibitory subunit of protein phosphatase 1 (PP-1A). Phosphorylation by PKC regulates its binding to PP-1A. Acts co-operatively to inhibit myosin light chain phosphatase (MLCP) activity. Inhibition leads to increased myosin phosphorylation and enhances smooth muscle contraction in the absence of increased intracellular Ca(2 ) concentration. Involved in the regulation of cell morphology, re-organisation of microfilaments, and cell spreading. Also expressed in human platelets and in brain where it may play a role in cerebellar long-term synaptic depression. Phosphorylation by ILK or Rho kinase enhances its ability to inhibit PP-1A.

Protein type: Protein phosphatase, regulatory subunit; Inhibitor

Chromosomal Location of Human Ortholog: 19q13.1

Cellular Component: cytoplasm

Molecular Function: protein phosphatase inhibitor activity

Biological Process: regulation of phosphorylation; negative regulation of catalytic activity
