Form/Format
Supplied in 100ul phosphate-buffered saline, 50% glycerol, 1 mg/ml BSA, and 0.05% sodium azide.
Purity/Purification
Affinity Purified
Specificity
The antibody was cross-absorbed to unphosphorylated Girdin (Tyr-1764) peptide before affinity purification using phospho-Girdin (Tyr-1764) peptide. This antibody detects a 250 kDa* protein on SDS-PAGE immunoblots of human A431 cells stimulated with EGF and Pervanadate. These reactivities are not observed after alkaline phosphatase treatment to dephosphorylate Girdin.
Preparation and Storage
Store at -20 degree C. Stable for 1 year.
Molecular Weight
250 kDa
OMIM

3D Structure
ModBase 3D Structure for Q3V6T2
Immunogen
Phospho-Girdin (Tyr-1764) synthetic peptide (coupled to carrier) corresponding to amino acid residues around tyrosine 1764 in human Girdin. This sequence has high homology to rat and mouse Girdin, and the site is not conserved in other Girdin family members.
Applications Tested/Suitable for anti-CCDC88A antibody
Western Blot (WB), ELISA (EIA)
Application Notes for anti-CCDC88A antibody
ELISA: 1:1000
WB: 1:1000
WB: Western blot membranes were incubated with diluted antibody in 5% non-fat milk, PBS, 0.04% Tween20 for 1 hour at room temperature.

Western Blot (WB) of anti-CCDC88A antibody

Western blot image of human A431 cell lysates treated with pervanadate (lanes 1-4). The blot was treated with alkaline phosphatase to dephosphorylate Girdin phosphosites (lanes 2 & 4). The blot was probed with mouse monoclonal anti-Girdin (lanes 1 & 2) or rabbit polyclonal anti-Girdin (Tyr-1764), phospho-specific (lanes 3 & 4).

NCBI Summary for CCDC88A

This gene encodes a member of the Girdin family of coiled-coil domain containing proteins. The encoded protein is an actin-binding protein that is activated by the serine/threonine kinase Akt and plays a role in cytoskeleton remodeling and cell migration. The encoded protein also enhances Akt signaling by mediating phosphoinositide 3-kinase (PI3K)-dependent activation of Akt by growth factor receptor tyrosine kinases and G protein-coupled receptors. Increased expression of this gene and phosphorylation of the encoded protein may play a role in cancer metastasis. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Dec 2011]

UniProt Comments for CCDC88A

girdin: a ubiquitously expressed actin binding protein that plays an important role in the formation of stress fibers and lamellipodia. Subsequent to phosphorylation by Akt, Girdin accumulates at the leading edge of migrating cells and appears to be crucial for the integrity of the actin cytoskeleton and for cell migration.
