Protein Function
Required for DNA double-strand breaks (DSBs) formation in unsynapsed regions during meiotic recombination. Probably acts by forming a complex with MEI4 and REC114, which activates DSBs formation in unsynapsed regions, an essential step to ensure completion of synapsis. Not required for HORMAD1 functions in pairing-independent synaptonemal complex formation, ATR recruitment to unsynapsed axes, meiotic silencing of unsynapsed chromatin (MSUC) or meiotic surveillance.
ISO Certification
Manufactured in an ISO 13485:2003 and EN ISO 13485:2012 Certified Laboratory.
Applications Tested/Suitable for anti-PPM1J antibody
Western Blot (WB), Immunocytochemistry (ICC), Immunofluorescence (IF), ELISA (EIA)
Application Notes for anti-PPM1J antibody
WB: 0.25-0.5ug/ml, Human, Mouse, Rat
ICC/IF: 5ug/ml, Human
ELISA: 0.1-0.5ug/ml

Western Blot (WB) of anti-PPM1J antibody

Figure 1. Western blot analysis of PPM1J using anti-PPM1J antibody (MBS1759455).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hacat whole cell lysates,
Lane 2: human MCF-7 whole cell lysates,
Lane 3: human U20S whole cell lysates,
Lane 4: human A431 whole cell lysates,
Lane 5: rat brain tissue lysates,
Lane 6: rat kidney tissue lysates,
Lane 7: mouse brain tissue lysates,
Lane 8: mouse kidney tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-PPM1J antigen affinity purified polyclonal antibody (#MBS1759455) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (#MBS176460) with Tanon 5200 system. A specific band was detected for PPM1J at approximately 55 kDa. The expected band size for PPM1J is at 55 kDa.

Immunofluorescence (IF) of anti-PPM1J antibody

Figure 2. IF analysis of PPM1J using anti-PPM1J antibody (MBS1759455).
PPM1J was detected in an immunocytochemical section of U2OS cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (#MBS176582) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5ug/mL rabbit anti-PPM1J Antibody (MBS1759455) overnight at 4 degree C. Cy3 Conjugated Goat Anti-Rabbit IgG (BA1032) was used as secondary antibody at 1:500 dilution and incubated for 30 minutes at 37 degree C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.
