Western Blot (WB) of anti-SYNGAP1 antibody

Figure 1. Western blot analysis of SYNGAP1 using anti-SYNGAP1 antibody (MBS1758537).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat brain tissue lysates,
Lane 2: mouse brain tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SYNGAP1 antigen affinity purified polyclonal antibody (#MBS1758537) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (#MBS176460) with Tanon 5200 system. A specific band was detected for SYNGAP1 at approximately 148 kDa. The expected band size for SYNGAP1 is at 148 kDa.

Flow Cytometry (FC/FACS) of anti-SYNGAP1 antibody

Figure 2. Flow Cytometry analysis of K562 cells using anti-SYNGAP1 antibody (MBS1758537).
Overlay histogram showing K562 cells stained with MBS1758537 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-SYNGAP1 Antibody (MBS1758537, 1ug/1x106 cells) for 30 min at 20 degree C. DyLight488 conjugated goat anti-rabbit IgG (MBS177405, 5-10ug/1x106 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

NCBI Summary for SYNGAP1

The protein encoded by this gene is a major component of the postsynaptic density (PSD), a group of proteins found associated with NMDA receptors at synapses. The encoded protein is phosphorylated by calmodulin-dependent protein kinase II and dephosphorylated by NMDA receptor activation. Defects in this gene are a cause of mental retardation autosomal dominant type 5 (MRD5). [provided by RefSeq, Dec 2009]

UniProt Comments for SYNGAP1

SynGAP: an excitatory synapse-specific RasGAP. Member of the NMDAR signaling complex in excitatory synapses it may play a role in NMDAR-dependent control of AMPAR potentiation and synaptic plasticity. ERK activation is up-regulated in neurons from SynGAP knockout mice, whereas P38 MAPK function is depressed. May play a critical role in the regulation of neuronal MAPK signaling, AMPAR membrane trafficking, and excitatory synaptic transmission. Interacts with CAMK2A/CAMK2B and MPDZ. Phosphorylated by CaM-kinase II. Dephosphorylated upon NMDA receptor activation or SYNGAP1/MPDZ complex disruption. Contains a PH domain and a Ras-GAP domain. Two alternatively spliced isoforms have been described.

Protein type: GAPs, Ras; GAPs

Chromosomal Location of Human Ortholog: 6p21.3
